Protein purification and working with enzymes are important components in a biochemistry lab course. I have students though out the term work with an assigned enzyme to research the enzyme’s physical properties using the biochemical literature, characterize the enzyme activity using kinetics assays and, finally, purify that enzyme using a purification scheme that the students have designed. These activities are done in addition to having students learn basic biochemical techniques for separating, quantifying and analyzing biochemical molecules. The students hand in a lab assignment over the literature research, a formal report over the enzyme kinetics experiments and an oral presentation over their purification protocols and outcomes.
